Join Dr. Glanville tomorrow October 22nd at 1:00 PM PST/ 4:00 PM EST for his presentation for Scripps Education: Immunology & Microbiology Bicoastal Seminar Series Webinar - "Although the novel pandemic SARS-CoV-2 was related to the previous SARS outbreak coronavirus, SARS-CoV-2 was sufficiently evolutionarily distinct that anti-SARS antibodies could no longer neutralize the novel virus. An analysis of the crystal structures of five neutralizing anti-SARS antibodies, drawn f...rom convalescent SARS patients, healthy controls, and murine immunizations, revealed that 28-75% of amino acids in their epitopes were not conserved in the novel coronavirus, affected 5-6 of contact CDRs in the respective antibodies. Being able to evolve these antibodies would be desirable, as over 4 years of crystal structures, neutralization and in-vivo protection validation had gone into these molecules, and evolving them while preserving their function would provide a rapid means to bypass a significant amount of engineering time for this as well as future pandemics, and also transition immediately from discovery into optimization of thermostability/affinity/humanness. Here we used a diversity compression/diversity superposition/computational design method to evolve these antibodies to affinity mature the antibodies against novel coronavirus in a single step while simultaneously optimizing multiple therapeutic characteristics. The results provide a therapeutic molecule for the acute need, but also clarify a general rapid strategy to repurpose characterized antibodies against homologous viruses while preserving their functions."
