A supporting cast serves the same purpose as any other element of a script: 1. To drive plot 2. To reveal character... How you weave this web and build relationships is up to you, but every interaction is an opportunity to reveal character and drive plot. Don' waste it. Each character should have their own role and desire in the script and actively pursue it. This creates more opportunities for conflict, tension, and subplots.

Make the antagonist more powerful than your protagonist and seemingly impossible to overcome. Think about the The Dark Knight Rises. Bane was seemingly unstoppable and literally destroyed Batman, which made Batman's trial and ultimate victory that much more satisfying.

In addition to the desire, the character must have an unconscious need. Action/horror films are somewhat of an exception. The need can be anything, but is strongest when based on a psychological or moral issue. They can only truly achieve their desire by overcoming their flaw.
